Summary:Free radical are molecules that have unpaired electron in their outer orbital, so they are very reactive and can attack healthy cells in the body. One of mecanism to reduce the free radical with antioxidant. Antioxidant is compounds can scavenge free radical or electron donor, so that the free radicals become more stable. Antioxidants can fight oxidation reaction by binding unpaired electron to be unreactive molecules and prevent damage. Antioxidant compound that commonly found from plant is flavonoid compound. One of the medicinal plants in Indonesia which has antioxidant activity is the crown of god (Phaleria macrocarpa (Scheff) Boerl. Flesh of crown of god have flavonoid compounds as antioxidant. The objectives of this research were to study antioxidant activity of leaves, branches and fruits extracts of crown of god by DPPH and FRAP methods, determine phenolic total and flavonoid total, analyze correlation between phenolic total and flavonoid total with their IC50 DPPH and EC50 FRAP and also analyze correlation between two methods. Extraction was performed by reflux using n-hexane, ethyl acetate and ethanol as solvents. Determination of IC50 DPPH, EC50 FRAP, phenolic total, and flavonoid total content each of extract were performed by spectrophotometry UV-visible. Correlation between flavonoid and phenolic content with their IC50 DPPH and EC50 ????????!????????±????± •?•?????????±­ ¢????????±•???????????????????±????í?­???? IC50 DPPH of leaves, branches and fruits extracts of crown of god in the range of 1.14 ????55.96 µg/mL and EC50 FRAP in the range of 33.33 ????338.85 µg/mL. The highest flavonoid and phenolic total were indicated by ethanol fruit extract of crown of god (17.45 ± 0.38 g QE/100 and 20.39 ± 0.36 g GAE/100 g extract). Generally leaves, branches and fruits extracts of crown of god was very strong antioxidant. Flavonoid compounds in fruit extract were main contributor in antioxidant activity by DPPH method. Phenolic compounds in branches extract were main contributor in antioxidant activity by FRAP method. While phenolic and flavonoid compounds of fruits extracts were joints contributions in antioxidant activity by FRAP method. DPPH and FRAP methods gave linear results in antioxidant activities of crown of god leaves and fruits extracts.
