Hence the use of extract from plants can be an alternative drug for treating insomnia; Water spinach?s leaves and gotu kola?s herbs have been used traditionally to reducing anxiety and helping someone to sleep. This study was aimed to formulate capsules containing a combination ethanol extract of water spinach?s leaves and gotu kola?s herbs ?????????:??????in order to obtain hypnotic activity with more effective and increased patient compliance. The crude drug powder of water spinach and gotu kola were extracted by maceration method using ethanol then concentrated into concentrated ethanol extract. Extract were formulated with various concentration of Avicel PH 102, lactose, aerosil, and magnesium stearat. Capsules mass were evaluated for organoleptic, flow test, and moisture content. The study of hypnotic activity using Swiss-Webster male mice that divided into 7 groups. The reference drug used was diazepam (3 mg/kg BW), 0.5% CMC-Na as vehicle group, and test group consist of formula A (306 mg/kg BW), formula C (306 mg/kg BW), formula E (200 mg/kg BW), and formula H (200 mg/kg BW) along with combination of water spinach and gotu kola ethanol extract group (100 mg/kg BW). The parameter measured were sleep latency, sleep duration, and sleep efficiency that statistically processed using One Way ANOVA and Post-Hoc Test (LSD). The result of this study shows that formula H which contain Avicel PH 102, aerosil 4%, and magnesium stearate 2% can be an option because it gave a good organoleptic, flow rate, and moisture content outcome among others formula. In addition, from evaluation of hypnotic effect, formula H had sleep efficiency of 89.99 ± 2.56% which was not significantly different (p<0,05) compared to extract combination (89.61 ± 1.65%) and diazepam (92.04 ± 0.77%). text
