Pharmacological effects of free radicals cannot affect human body directly, but accumulation from free radicals can causes distruption of human body’s mechanism. Antioxidant are able to prevent oxidation process that caused by free radicals. Antioxidants can be obtained from plants, one of them is okra (Abelmoschus eschulentus L. Moench). The objective of this research were to study antioxidant activity from branches, leaves, and fruits extract of okra by determining EC50 CUPRAC and IC50 DPPH; determine total phenolic and flavonoid content analyze the correlation between total phenolic and total flavonoid with EC50 CUPRAC dan IC50 DPPH; and analyze the correlation between CUPRAC and DPPH method in sample extracts. Determination of EC50 CUPRAC dan IC50 DPPH, total phenolic and total flavonoid content of each extract were performed by UV-Visible spectrophotometry. The correlation of total phenolic and total flavonoid content with their EC50 CUPRAC dan IC50 DPPH and also correlation of two methods were analyzed using Pearson method. Antioxidant activities test results of branches, leaves, and fruits extracts of okra gave IC50 CUPRAC in the range of 2,240 – 24.059 µg/mL. The highest total phenolic was given by the ethanol leaves extract of okra (3.198 ± 0.016 g GAE/ 100 g ) and the highest total flavonoid content was given by ethyl acetate extract of okra (4.279 ± 0.201g QE/ 100 g ).Total phenolic content had negative and significant correlation with their EC50 CUPRAC and IC50 DPPH in all sample extracts. All of the organ extract like branches, leaves, and fruits were categorized as a very strong antioxidant by DPPH method, so they have potential and can be considered to be developed as sources for natural antioxidants. CUPRAC and DPPH method did not showed any linear results in measuring antioxidant activity from all of the sample. text
