Summary:Plectranthus scutellarioides (red jawer kotok leaves) is a decorative plants, which traditionally used to treat various diseases. Jawer kotok leaves contains essential oil, diterpene and flavonoid, but the research has not been reported on spesific chemical content of this plants. This research was carried out to analyze chemical content from red jawer kotok leaves and the result hopefully could be used to identification of the plant. Isolation of flavonoid performed by extraction of red jawer kotok leaves with maceration method using ethanol 96%. The extract was then fractionated by the Charaux-Paris modified method using n-hexane, chloroform, ethyl acetate and n-butanol solvent. The content of each fraction was identified by Thin Layer Chromatography (TLC). The selected fraction was separated by classical column chromatography and purification was carried out by recrystallization that obtained isolate A. TLC using three different polarities mobile phase and isocratic High Performance Liquid Chromatography (HPLC) using metanol-acetic acid 5% (6 : 4) as mobile phase were used to identify the purity of isolate. The isolate were characterized by UV-Visible Spectrophotometry and Nuclear Magnetic Resonance (NMR-1H). Phenolic acid obtained from ethanol extract by Liquid-Liquid Extraction (LLE) method using 3 procedure, that was without hydrolysis, acid hydrolysis and alkaline hydrolysis. Then, each extract were identified by TLC with phenolic acid standards using the Reverse Phase-18 F254S as stationary phase and methanol-water (6:4) as mobile phase. Identification of phenolic acid was carried out by looking at the spots in the extract which had the same color and Rf as the standard compound, then compared the UV spectrum of each spot using TLC Scanner. The result of the analysis one of the flavonoid compound in red jawer kotok leaves was 5,7,3’,4’-tetrahydroxyflavone (Luteolin), and one of the phenolic acid in red jawer kotok leaves was predicted as ferulic acid.
