Summary:Endophytic fungi are fungi that are associated with their host's internal tissue. Some endophytic fungi can stimulate the production of secondary metabolites in their hosts that have activities as antimicrobials. Endophytic fungi could be found on many plants, such as mangrove plants (Rhizophora sp.). The aim of this reseach is to isolate endophytic fungi from mangrove roots originating from Sei Sembilang village, and to test the antimicrobial activity of endophytic fungi on Escherichia coli (ATCC 8739), Staphylococcus aureus subsp. aureus (ATCC 6538), and Candida albicans (ATCC 10231). This research consisted of mangrove root sampling, isolation and purification of endophytic fungi, fermentation and extraction of secondary metabolite, antimicrobial activity screening tests with disc diffusion method, determination MIC by microdilution method, and determination of MBC/MFC by growth method on agar media. The research obtained 10 pure endophytic fungi isolates with 20 types of test extracts. The results of the antimicrobial activity screening test obtained that 18 extracts that had activity against Escherichia coli, 14 extracts had activity against Staphylococcus aureus and 8 extracts had activity against Candida albicans. The extract with the lowest MIC value against Escherichia coli is medium extract of FDSS-02 (32 ?g/mL), against Staphylococcus aureus is medium and mycelium extract of FDSS-02 (64 ?g/mL), and against Candida albicans is medium and mycelium extract of FDSS-2 (64 ?g/mL). The extract with the lowest MBC value against Escherichia coli is a medium extract of FDSS-02 (32 ?g/mL), against Staphylococcus aureus is mycelium extract of FDSS-01, FDSS-08, FDSS09, and FDSS-10 (256 µg/mL).The lowest MFC value against Candida albicans is medium and mycelium extract of FDSS-02 (64 ?g/mL). This research has potential to discover a new antimicrobial sources.
