Summary:Sow thistle (Sonchus arvensis Linn.), roselle (Hibiscus sabdariffa Linn.), and madeira vine (Anredera cordifolia (Ten.) Steenis) were traditionally used as treatments for lowering blood uric acid. These three plants contained many secondary metabolite compunds which had potential effect as xanthine oxidase inhibitor. The aim of this study was to determine IC50 value of sow thistle leaves extract (B), roselle calyces extract (R), madeira vine leaves extract (T) and their combination (1:1), also the characteristic of those combinations. In this study, a phytochemical screening and in vitro xantin oxidase inhibition assay were conducted on single-extract, combination extract (1:1) and the allopurinol as a compararitor drug. Phytochemical test of extract T showed the presence of alkaloid, steroid/triterpenoid, flavonoid, tannin and saponin. Meanwhile extract R showed the presence of steroid/triterpenoid, flavonoid, saponin, dan quinone and extract B only showed the presence of steroid/triterpenoid, flavonoid, and saponin. IC50 of extract T, R, B were 1165.045; 3122.730; 3158.235 µg/mL respectively. Extract T had bigger inhibition of xanthine oxidase than extract R and B. Allopurinol showed highest inhibitory activity on xanthine oxidase than all of the extract with IC50 0.97 µg/mL. Combination of extract T-R, T-B, R-B showed that they had an anti-gout effect but combination of extract T-R was the gretest combination of all with IC50 1805,461 µg/mL. Combination of extract T-R and T-B might have an additive effect while the combination of extract R-B showed a synergistic effect.
