Summary:Kaolin is a natural hydrated aluminum silicate which was usually combined with pectin, a polysaccharide, used as antidiarrheal because it has activity as an adsorbent. Aluminum content in kaolin was determined by the spectrofluorometry method through derivatization reaction using 8- hydroxyquinoline reagent at pH 6 in chloroform solvent. Fluorescence intensity measurements were carried out at the maximum excitation wavelength of 384 nm and maximum emission wavelength of 525 nm. Validation method was done by determining linearity, accuracy, precision, detection limit, and quantification limit. The validated method was tested to simulations of kaolin-pectin tablet powder and suspension. Dry method destruction of the simulation sample was carried out at a temperature of 800 o C for 5 hours. The residue was dissolved in sulphuric acid and the filtrate was taken to be derivated. Determination method of aluminum content in kaolin with spectrofluorometry has been validated with linearity as correlation coefficient (r) of 0.9979 and variance regression coefficient (Vxo) of 3.79%; detection limit of 0.23 ppm and quantification limit of 0.76 ppm; accuracy test was performed to tablet powder and suspension simulations, resulted in %recovery of 92.36±0.49%-102.61±1.58% and 97.22±0.57%-105.83±0.64%, respectively; and interday precision test to tablet powder and suspension simulations, resulted in Horwitz Ratio (HORRAT) value of 0.06-0.07 with SBR 0.85%-1.01% and HORRAT 0.09-0.13 with SBR 1.25%-1.74%, respectively, with each value meet the requirements based on the AOAC International Guideline, a guideline for validating analytical methods. The content of aluminum contained in kaolin was 21.15±8,85 ppm. This validated method was applicated to define content of Tablet “O”with 12,249 ppm ± 3,684% of aluminium and 579,084 mg ± 3,684% of kaolin in each tablet.
