Summary:Cassava flour or usually called fercaf is cassava modified through fermentation process that could improve the quality of cassava itself. Fermentation process of cassava needed microbial starter as fermentation agent, the microbial starter that used for fercaf are Aspergillus oryzae and Bacillus subtilis. The more fercaf flour’s demand, the more microbial starter too. This research has purpose to finding out the proper method to producing starter microbial flour in pilot scale. This experiment has three variations, those are rasio of medium substrate and inoculum, adding macronutrien in substrate, drying temperature process and evaluation of scale up process in tray bioreactor. The experimental results of microbes viability will be analyze using Total Plate Count method and others analyze i.e, water activity, CO2 analyzer, moisture content and relative humidity. Result of this research indicate that rasio medium substrate and inoculum 1:1 gives the best viability both in fermentation and drying process for Aspergillus oryzae and Bacillus substilis, i.e 3,39 x 106 cfu dan 13,06 x 1010 cfu after drying. Macronutrient adding in fermentation process is effecting growth of both microbes colonies significantly, but variation of three type macronutrient those are combine of K2HPO4, Al2(SO4)3 dan (NH4)2SO4 not different significantly. In drying process, temperature 50oC gives the more effective from microbe’s viability and water activity in the last process that can achieve Aw value until ±0,1. Scale up process in this research show increasing colony of microbes significantly, i.e 2 more for Aspergillus oryzae in addition of N and S, also 10 more for Bacillus substilis. The system of scale up process using tray bioreactor is considered have good enough based from homogeneity of moisture content and water activity’s material in each tray.
