A Genome (Musa acuminata) and B Genome (Musa balbisiana) bananas are the parents of most banana cultivars in Indonesia. One of banana’s nutritional content is beta-carotene, and its content in a fruit is determined by genes regarding its degradation. One of beta-carotene’s degradation pathway is its conversion to strigolactone by the enzymes: beta-carotene isomerase (D27), carotenoid cleavage dioxygenase 7 (CCD7), and CCD8. In this research, the characterization of those genes’ structure in M. acuminata ‘DH-Pahang’ and M. balbisiana ‘PKW’ (Pisang Klutuk Wulung) was done, along with identification of domains and motifs in D27, CCD7, and CCD8 putative proteins. Gene sequences, gene structures, and putative proteins of CCD7, CCD8, and D27 in DH-Pahang were retrieved from GeneBank Meanwhile, PKW’s genomic DNA was retrieved from Banana Genome Hub and its gene sequences were predicted based on the genes of DH-Pahang. Characterization of nucleotide structure included sequence length and numbers and position of exons and introns of each genes in DH-Pahang and PKW, analysis of similarity percentage between the same gene sequences in the two cultivars with pairwise sequence alignment, and motif and domain identification from each putative protein sequences. Phylogenetic tree construction for CCD7 and CCD8 putative proteins was done along with other CCD proteins of DH-Pahang and Arabidopsis thaliana; also with the same protein sequences from various plants. D27, CCD7, and CCD8 genes in DH-Pahang and PKW have various nucleotide sequence length, putative proteins, and numbers and length of exons and introns. Despite that, the putative proteins have the same domains: DUF4033 (Domain of Unknown Function) in D27 and RPE65 (Retinal Pigment Epithelial membrane) in CCD7 and CCD8. Phylogenetic trees produced showed that D27, CCD7, and CCD8 proteins from DH-Pahang and PKW are conserved and clustered in the same clades with the same proteins of monocot plants. text
