Silica amine (Si-NH2 ) column is commonly used in HPLC for sugar analysis, however, it is considered expensive for a column. In this research, a Si-NH2 column has been made with silica particles with the size of 30-60 micron which is being modified with 2% and 20% (3-Aminopropyl)trimethoxysilane (3-APTMS). The first step in this experiment begin with activating the silica particles using piranha solution made up of H so. an d 1-1 2 0 30% (3: I). The activated silica is then reacted with 3-APTMS for 24 hours in order to obtain Si-NJ-12 particles, and some of the Si-NH 2 particles go through deactivating process with triethylamine. Every step is followed by characterization using FT-IR and at the end SEM analysis is done. According to FT-IR data, Si-NJ-12 particles are successfully made due to new peaks fanned on the spectrum such as peaks from Si-C (800-950 cm- 1 ), C-N (1000-1350 cm- 1 ), dan N-1-1 ( 1550-1650 cm-1 ). Afterwards, Si-NJ-1 particles are loaded to HPLC column with size of 4.6 mm x I 00 mm. Column is being tested upon glucose, maltose, and a mixture of both which are made by three different solvents similar to the eluents used. Eluents used involves water, aceton itrile, and water-acetonitrile (50:50 ). Based on the experiment, Si-NJ-12 made with 2% 3-APTMS (Si -NJ-12 2%) with eluent of water-acetonitrile (50:50 ) gives the best perfonnance where glucose and maltose sample peaks are formed on different retention time with significant peak shapes.
