Cryptocarya plants produce matn secondary metabolites which belong to flavonoid, pyrone, and alkaloid types having various bioactivities, such as anticancer, antiplasmodial and analgesic. One of the species belonging to Cryptocarya genus is C. tomentosa. There were previous researches on this species, i.e. two researches from Indonesia studied the leaves of C. tomentosa and the bark of C. ferrea, which is C. tomentosa's botanical synonym, while another research from Malaysia studied the bark of C. ferrea. From the leaves of C. tomentosa , four flavonoids were found identitied as quercetin, dihydroquercetin, (+)-catechin, and taxifolin-3-0-a-L-arabinofuranoside. From the bark of Indonesian C. ferrea, one flavonoid and two steroids were found , i.e. (-)-pinocembrin, P-sitosterol, dan stigmasterol whereas from the bark of Malaysian C ferrea, three alkaloids were found namely as (-)-0-methylisopiline, (+)-norlirioferine, and (+)-lirioferine. In this research, secondary metabolites have been isolated from the bark of C. wmentosa. The stembarks were obtained from Bogor Botanical Gardens, West Java. The isolation of compounds was conducted by several steps including maceration of stembark powder with acetone, then fractionation by vacuum liquid chromatography, and purified further by several chromatography tecniques, such as gravitational colur:m chromatography and radial chromatography. In this experiment, three isolates have been obtained which were subsequently analyzed using one dimensional N1\1R spectroscopy ctH-NMR and 13C-NMR) and two dimensional NMR spectroscopy (HSQC and HMBC). One of the isolate was identified as (-)-pinocembrin and two others contained major compound analyzed as (±)-chartaceone A and (+)-catechin, respectively. The finding of chartaceone A is the first time from Indonesian Cryptocary:J. It was discovered previously from the stem bark of C. chartacea grown in New Caledonia. Furthermore, the cytotoxicity test against murine leukemia cell line P-388 showed that the extract was active (lCso = 7.91 Jlg/mL), whereas (-)-pinocembrin (IC50 = 17.3 w'mL) and the isolate containing chartaceone A (IC5o = 6.08 ?/mL) were not active. text |
