Bacteria in bioaerosol have been found to have a relationship with infectious and non-infectious diseases in humans that affect air quality. In this study, analysis of the air bacterial community at Tanjung Priok Port, Jakarta City, was carried out to determine the amount and diversity of bacteria found in that place. Air sampling was carried out using durapore membrane filter (0.5 micron) which was then used for analysis. Air sampling was carried out using the durapore filter membrane (0.5 micron) which was then used for analysis with a culture-dependent and culture-independent approach. The culture-dependent analysis consisted of measurements of bacterial abundance by the total plate count method on the Nutrient Agar, Tryptic Soy Agar, and Luria Bertani medium, and the determination of the bacteria diversity by the process of sequencing by Macrogen. Analysis with culture independent method was carried out by extracting DNA from membrane filter which was then followed by the sequencing process using Oxford Nanopore Technologies MinION. Measurements of endotoxin concentration also caarried in air samples using the chromogenic LAL test (Limulus Amebocyte Lysate).The results showed that bacterial abundance in NA, TSA and LB medium respectively is, 2.50 x 107 CFU/m3; 3.37 x 107 CFU/m3; and 3.00 x 107 CFU/m3 during the day. Whereas at night the bacterial abundance is 7.40 x 106 CFU/m3; 1.06 x 107 CFU/m3; and 8.80 x 106 CFU/m3. The diversity of bacteria found by culture-dependent methods consists of Bacillus subtilis included in the phylum Firmicutes, Pseudomonas stutzeri which is included in the phylum of Proteobacteria, Kocuria palustris and Isoptericola nanjingensis which are included in the phylum Actinobacteria. Whereas the results of the analysis using the culture-independent method show that the air of the Tanjung Priok Port in Jakarta is dominated by phylum Proteobacteria at day and night. Bacterial species that are potentially pathogenic to humans are also found at the sampling site. During the daytime species of bacteria that have potential pathogens were found, among others, Escherchia coli (38%), Salmonella enterica (4.9%), and Shigella flexneri (1.4%) whereas at night Delftia acidovorans (10%) , Escherchia coli (4%) and Delftia lacutris (4.4%), was found. The concentration of endotoxin in the Tanjung Priok Port air is detected at 17.97 EU / m3 during the day and 35.19 EU/m3 at night, while the concentration that is safe for health is 30.00 EU/m3. So that bacterial diversity and endotoxin concentration in this study show that air quality at Tanjung Priok Port in Jakarta City has the potential to cause infection. text
