Actinobacteria has a high level of productivity in producing bioactive compounds. The potential of endophytic Actinobacteria in soft corals has not been widely explored. The vulnerable condition of an individual contracting a bacterial infection, the potential of bioactive compound as antibacterial agent is needed to be explored further. Actinobacteria was isolated from soft coral extract by dilution method using Yeast Malt Agar (YMA) medium. Antibacterial potential was carried out through antagonistic test using dual-culture methode in E. coli, B. subtilis, and S. aureus cultures. The Actinobacteria that showed the highest inhibition were further selected in the second screening using Kirby-Bauer method. The best Actinobacteria that showed the best antibacterial activity then be tested to find it’s antibacterial potential by comparing the diameter of inhibition zone between Actinobacteria extract in one dose with universal antibiotic in five doses. The universal antibiotic was streptomycin. Doses that use in comparation are 30 ppm for Actinobacteria extract and 19,2 ppm, 24 ppm, 30 ppm, 37 ppm and 46,8 ppm for streptomycin. Soft coral that have been founded in the litoral zone at Rancabuaya beach are Haliclona cf. baeri, Haliclona caerulea, Clona verde and Spheciospongia vagabunda. Four Actinobacteria that was successfully isolated from these marine soft corals and show antibacterial activities were Streptomyces katrae, Streptomyces galbus, Streptomyces violaceus and Saccharopolyspora antimicrobica. Saccharopolyspora antimicrobica was the best endophytic Actinobacteria in Haliclona cf. baeri as an antibacterial agent with the inhibition zone of 16,83 mm in culture of S. aureus and 19,26 mm in culture of B. subtilis. The value of the antibacterial potential test showed that Saccharopolyspora antimicrobica had 95,5% potential against B. subtilis and 84,45% against S. aureus. text |
