This plant is known to have secondary metabolites, such as saponins which accumulate in the root organs. Saponin itself acts as an anti-inflammatory agent, increasing fertility and endurance. In nature, Javanese ginseng takes a very long time to produce saponin optimally, so in vitro of root culture techniques are needed to increase total saponin production in Javanese ginseng. This study was conducted to investigate the effect of immersion duration on the growth of adventitious root and total saponnin content. This study was conducted using 3 different immersion durations in the same interval (15 minutes, 30 minutes and 60 minutes immersion with 3 hours interval). Explants were grown on Murashige and Skoog (MS) medium with the addition of IBA (indole-3-butyric acid) and NAA (1-Naphthalene acetic acid) hormones. Roots grown on IBA 10 ?M hormones produce the highest growth compared to the others (0,035 g/day of growth rate). This optimum consentration was used for adventitious root incubation in ½ MS liquid medium in TIS RITA® bioreactor with different immersion duration. Culture with 30 minutes immersion duration produced higher biomass than others (0.0126 g/day), while the highest saponin content was obtained in culture with 15 minutes immersion time (3,48%). Based on these results, adventitious root culture uses a TIS RITA® bioreactor with a 15 minutes immersion duration at 3 hour intervals is better in saponin production in Javanese ginseng. text
