This study focused on increasing the accumulation of gallic acid in Spirogyra sp. by applying various photoperiodism, which was 8 hour light 16 hour dark, 12 hour light 12 hour dark, 16 hour light 8 hour dark, and 24 hour light 0 hour dark in Bold Basal Medium (BBM) 2,5% strength. Cultivation last for 21 days in plastic container with dimensions 16 x 8 x 7,5 cm filled with 500 mL BBM. Sampel taken every 3 days with 3 repetition for each data for testing. The analysis of gallic acid was performed by using High Performance Liquid Chromatography (HPLC). The HPLC conditions needed in this study were 18C Column, combination of 2 eluent: methanol HPLC grade (A) and H3PO4:H2O = 0,05% (B) with composition of A:B = 90:10 (10 minutes); 70:30 (3 minutes); 40:60 (5 minutes); 60:40 (3 minutes); 80:20 (3 minutes); and 90:10 (6 minutes), flow rate 1 mL/minute, and detector UV-Vis at 271 nm. Results indicated that the highest accumulation of gallic acid obtained in 16 hour light photoperiodism with total value of 17,77 mg/g or 1,78% from its dry weight. 16 hour light photoperiodism increased the total gallic acid accumulation by 2,14 times from 12 hour light variations. Highest gallic acid productivity estimation from Spirogyra sp. calculated by using the data from 16 hour light and 12 hour light photoperiodism and give a total of 864,702 kg gallic acid/year/hectare for 16 hour light and 757,2104 kg gallic acid/year/hectare for 12 hour light photoperiodism. text |
