Summary:Astaxanthin is a ?-carotene derivative compound with high economic value, because its has very strong antioxidant ability. Astaxanthin is reported produced by fresh water macroalgae Spirogyra sp.. Astaxanthin accumulation from several types of green algae can be increased under oxidative stress condition, one of which through the induction of iron ion (Fe2+). The aim of this study was to increase accumulation of astaxanthin in Spirogyra sp. by induce iron ion (Fe2+) to the culture of Spirogyra sp.. Initial cultivation was conducted to optimize the biomass of Spirogyra sp. that cultivated in two different medium, manure medium 0,5% (w/v) and Bold Bassal Medium (BBM) 2,5%. Cultivation was conducted for 14 days in transparent container 16 x 16 x 7,5 cm, illuminated 2500-300 by flourescens, aerated by ambient air 1 vvm, and cultivation condition was kept at 25±1oC and pH 7.2-7.8. Mathematical model of kinetic growth was constructed to estimate kinetic growth parameter and determine the optimum medium for Fe2+ induced-stage cultivation. The addition of Fe2+ was conduct on the 6th day of Spirogyra sp. that cultivated in BBM 2,5%. Concentration Fe2+ has been varied to 50 ?M, 75 ?M, 100 ?M, and control. Spirogyra sp. was harvested every two days using vacuum pump and fresh culture was dried with freeze drying method. Astaxanthin content was determined by extracted the dried biomass with maceration method using methanol-dicloromathane (1:3,v/v) for 48 hours. Astaxanthin content was analyzed using High Performance Liquid Chromatography (HPLC) with a 18C column and mobile phase methanol-H2O (95:5). Highest astaksantin accumulation reached 0,208 mg/g with Fe2+ 100 ?M on the 8th day, increased 3,3 times of control. However, the result of biomass growth of Spirogyra sp. with addition Fe2+ lower than control culture. The estimated productivity of astaxanthin in Spirogyra sp. with addition Fe2+ 100 ?M reached 9,117 kg/hectare.year.
