Gallic acid accumulation in Spirogyra sp. is known to be enhanced through administration of Cu2+ stress, which can provide oxidative stress through reactions similar to the fenton reaction thereby activating the shikimic acid pathway, the pathway to synthesize gallic acid. In this experiment, stress was given by varying the concentrations of Cu2+ in the medium, which is below normal (0.0785 ?M), normal (0.157 ?M), and above normal (0.2355 ?M and 0.314 ?M). Spirogyra sp. biomass was harvested every three days and its fresh weight was, then dried using freeze-drier. Dry biomass of Spirogyra sp. was extracted with methanol. Gallic acid levels were analyzed using High Performance Liquid Chromatography (HPLC) with UV-Vis detector at ?= 271 nm and C18 column. The highest yield of gallic acid was obtained at a concentration of Cu2+ 0,157 ?M or without stress, which was 8,303 mg/g. Estimated productivity of gallic acid on an industrial scale is 757,21 kg / ha / year. text
