In Indonesia, research about the regenerative potential of conditioned medium from umbilical cord-derived mesenchymal stem cell for wound healing purpose, has been carried out. Lyophilization is a method that can be used to stabilize conditioned medium, which contains complex mixture of protein. In order to minimize protein degradation during lyophilization, lyoprotectant needs to be added. The aim of this study is to evaluate the effect of type and quantity variation of lyoprotectant on physical and biochemical attributes of lyophilized conditioned medium. Four types of lyoprotectant, mannitol, trehalose, glycine, and sucrose, were used in this study. Concentration for each lyoprotectant was 0,5, 3,0, and 7,0% (m/v). Physical evaluation consists of cake appearance and morphology, as well as appearance, pH, and osmolality of solution before lyophilization and after reconstitution. Biochemical evaluation was carried out for Human Pro-Collagen assay in conditioned medium. The study shows that glycine and sucrose are prospective to be used as lyoprotectant in lyophilized conditioned medium from human umbilical cord-derived mesenchymal stem cell. Furthermore, optimum result can be observed in 3,0% glycine concentration. This concentration shows high preservation of Human Pro-Collagen as well as suitable appearance, pH, and osmolality for subcutaneous injection dosage form. text
