Summary:Nanoparticles uses for drug delivery system of as-ODN as malaria therapy agent are to prevent enzymatic degradation and to improve as-ODN deliverance to target. Freeze dried is performed to nanoparticles preparation with the purpose is to improve the efficiency of packaging and use of preparation. Cryoprotectant is added to nanoparticles to prevent freezing and drying stress while nanoparticles is performing freeze dry. The purpose of this study is to find the optimum cryoprotectant that could maintain the particle’s diameter of nanoparticles while freeze dry processing. Optimization of nanoparticles was performed to mannitol, lactose, glucose, fructose and dextran with 1%, 5%, 10%, 15% and 20% concentrations. The optimum cryoprotectant was assessed by ratio of nanoparticles diameter before and after freeze dry, the closer the ratio to 1 the better. The optimum cryoprotectant to protect freezing and drying stress are 15% lactose and 10% glucose for chitosan nanoparticles formulation and 10% glucose and 10% mannitol for chitosan-poloxamer nanoparticles. Freeze dried chitosan nanoparticles has diameter particles, polydispersity index and zeta potential: 115,97-246,83 nm, 0,209-0,457, and +7,84 until +26,00 mV respectively. While freeze dried chitosan-poloxamer nanoparticles has diameter particles, polydispersity index and zeta potential: 161,5-212,7 nm, 0,335-0,451, and -1,80 until +15,40 mV respectively. Reconstitution time of freeze dried nanoparticles was between 71,5-832,5 seconds. Thermal analysis showed that there are some new peaks at thermal curve of freeze dried nanoparticles with cryoprotectant than freeze dried nanoparticles without cryoprotectant. Morphology observation of freeze dried product with SEM showed the difference between freeze dried nanoparticles before and after cryoprotectant addition.
