To overcome the issue of low immune response, in the present study, we attempted to enhance it by loading Phyllanthus niruri (P. niruri) L. along with HBsAg into nanoparticles. Nanoparticles were prepared by ionic gelation method using chitosan polymer and sodium tripolyphosphate as cross linker. Concentration of HBsAg used was 10 µg/mL while that of P. niruri extract used was 150, 250 and 350 µg/mL, respectively. The particles were characterized by size, zeta potential, entrapment efficiency, and the release of HBsAg. In vitro dendritic cells maturation study was conducted by treating immature dendritic cells by the nanoparticles. The percentage of dendritic cell maturation was determined by measuring the intensity of CD40, CD83 and CD86 with flowcytometry as mature dendritic cell marker molecule. Nanoparticles were having a size of 180-300 nm and a positive zeta potential of 27-32 mV. Entrapment efficiency study showed that 83-91% HBsAg and 61-84% P. niruri extract were entrapped in nanoparticles. In vitro released study exhibited that more than 90% HBsAg and P. niruri exctract were released within 2 hours. Furthermore, maturation study showed that HBsAg-P. niruri loaded nanoparticles has significant maturation (p<0,05) compared with that of HBsAg nanoparticles in concentrations of P. niruri extracts 250 and 350 µg/mL and costimulatory molecules used. Nanoparticles loading HBsAg and P. niruri were a potential carrier to induce maturation dendritic cells. text
