Uric acid is the result of purine catabolism which is aided by guanase and xanthine oxidase enzymes. The xanthine oxidase enzyme will catalyze the reaction of hypoxanthine to xanthine and the reaction of xanthine to uric acid. In Indonesia, many plants have the potential as xanthine oxidase inhibitors and they are effective in reducing uric acid levels, so this research will examine the inhibitory activity of xanthine oxidase enzymes of 8 ethanol extract from the Piperaceae family. From the selected extract was continued to fractionation to get IC50 value. The principle of measuring xanthine oxidase inhibitory activity is based on the amount of uric acid formed in the reaction catalyzed by xanthine oxidase and was expressed in percent (%) inhibition. Measurement using a UV-Vis spectrophotometer at a wavelength of 290 nm. The results showed that the eight ethanol extracts of the Piperaceae family had xanthine oxidase inhibior activity with an inhibition range of 6.75– 68.39% at a concentration of 100 µg/mL. Ethanol extract of betel leaf (Piper betle L.) had the highest xanthine oxidase inhibition activity, which was 68.39 ± 0.76%. The IC50 value of ethanol extract, ethyl acetate fraction, n-hexane fraction, and water fraction from betel leaf were 64.69 ± 0.48 µg/mL; 2.96 ± 0.12 µg/mL; 44.43 ± 0.38 µg/mL; and 955.33 ± 7.40 µg/mL. Meanwhile, the IC50 value of allopurinol as a positive control was 0.96 ± 0.06 µg/mL. This research showed that the use of the Piperaceae family has potential as an xanthine oxidase inhibitor, especially betel leaf (Piper betle L.). text
