Summary:Liver is one of the most important organ in the body that plays a main role in metabolism. Liver damage can caused by many factors, such as parasite infection, autoimmune disease, side effect of drugs such as antituberculosis and overconsumption of alcohol. The purpose of this study is to evaluate hepatoprotective activity of moringa leaves extract in male Wistar rats. Active substances on moringa leaves is extracted by reflux using ethyl acetate. Moringa extract was also formulated in nanosuspension dosage form with SLS & PVP as stabilizer. Through stability test at three different temperature and freeze & thaw, the most stable nanosuspension was obtained using SLS 0.5% as stabilizer. Hepatoprotection effect was studied through preventive and curative test with fixed dose combination of antituberculosis drug i.e. isoniazid (27 mg/kg BW) and rifampicin (54 mg/ kg BW) as inducer given intraperitoneally. The two doses of extract (75 & 150 mg/kg BW) tested either in crude extract or nanosuspension formed showed hepatoprotective effect indicated from GPT,GOT, ALP, GGT, and total bilirubin activity which is significantly lower (p<0.01) compared to than that of sick group. Moringa extract in nanosuspension formulation has better curative effect shown by significantly lower of GPT, GOT, ALP, GGT, and total bilirubin activities (p<0.05) compared to moringa extract which is only decreasing GPT, GOT, ALP activities (p<0.05). Liver tissues microscopic examination showed moringa extract in nanosuspension formulation could repair liver damage faster compared to moringa extract. Based on this study, moringa extract which is prepared in nanosuspension form was more potent compared to moringa extract as hepatoprotector.
