Summary: | Artemether is a potent derivative compound of artemisinin that have effect as antimalarial. Artemether
has limitations especially solubility in water and instability in acidic conditions, consequence the
bioavaibility of the artemether is low (40%). The low bioavalability could be improved by formulating
the artemether loading SLN (Solid Lipid Nanoparticles). The purpose of this research is to determine
the best formulation of SLN loaded artemether by screening suitable type of lipid, stabilizer and
determining concentration of active substance furthermore optimazing of the preparation process. The
method used in preparation of SLN was combination of high speed homogenization and
ultrasonication. The characteristic of the SLN was evaluated through organoleptic test, zeta potential,
particle size and morphology, physical and chemical stability test, drug entrapment efficiency, and in
vitro release test of the active substance. Based on the optimization process, the best velocity of
ultraturrax was 12000 RPM with duration of ultraturrax of 13 minutes, and duration of sonication of 1
minute. The best formulation of SLN for artemether consisted of artemether (0,75%); Precirol (6%); and
cremophor RH40 (7,5%) with particle size of 227 nm. The SLN was physically and chemically stable after
being stored at room temperatures until 7 days. Obtained SLN with particles size of 227 nm provided
entrapment efficiency of 87,37 ± 0,15%. The morphology of the particles was almost spherical with
particle size between 200-250 nm. According to in vitro release test, the active ingredient was released
almost 90% after 5 hours in phosphate buffer media pH 7,4. Artemether was successfully formulated
into SLN and stable until 7 days of storage.
