Summary:Natural ingredients are great potencies commonly used in pharmaceutical product especially in Indonesia. One of the plants which contains various natural active compounds used in Indonesia is Gnetum gnemon with the local name Melinjo which is the original product of Southeast Asia and some other countries in the world. Previous study has shown that melinjo seed extract has an effect decreasing uric acid on Japanese male. Decreased levels of uric acid is an attempt to overcome hyperuricemia. Uric acid is the final product of purine metabolism catalyzed by xanthine oxidase. This study aimed to know the activity of melinjo hard shell seed extract, and to evaluate the influence of particle size of melinjo hard shell seed extract on inhibitory effect of xanthin oxidase performed in vitro. The extraction was done by 50% ethanol maseration method and fractionation was done using HPD 600 resin. Size reduction of fraction collected was done by using top down technique which is ball milling. The result of nanoparticle is charaterized by size, morphology, and inhibitory activity performed in vitro. Scanning Electron Microscope showed differentiation between fraction morphology and nanoparticle melinjo hard shell seed extract morphology and the formation of particle in nanoscale. The inhibitory effect of extracts, fraction and the nanoparticle was concentration-dependence. At concentrations of 100 ppm; 200 ppm; and 400 ppm, the percentage of inhibition of extract, fraction, nanoparticles milling were respectively 24,94%, 54,84%, 70,72%; were 43,91%, 71,27%, 87,64%; and 67,05%, 89,25%, 92,23%. In conclusion, melinjo hard shell seed extract has xanthin oxidase inhibitory activity then formation of nanoparticle increased the inhibitory activity.
