Summary:Antibiotics resistence is one of many serious health problems that emerged both in developed and advanced countries in the world. One of many examples of antibiotics resistance is Mycobacterium tuberculosis resistence to standard therapy for tuberculosis. Alternatives treatments have been done to overcome Mycobacterium tuberculosis resistence. Developing drugs from natural product is one of solutions. Pomegranate fruit (Punica granatum L) has been used traditionally as an antimicrobial agent. This research was done to determine LC50 of the extract, interactional studies between the subtances that contained in pomegranate peel extract with enzym in Mycobacterium tuberculosis, and to predict its toxicity. Fish embryo acute toxicity was done by using zebrafish (Danio rerio) embryo as subject to obtained safety characteristics of extract. Pomegranate peel ethanol extract had LC50 196,037 ± 9,2 µg/mL. Based on aquatic toxicity classification by OECD, pomegranate peel ethanol extract was classified as safe. Interactional study was done to determine activity of pomegranate ethanol by docked the subtances with MTB enoyl acil carrier protein reductase (PDB ID : 2H7I), MTB Gyrase B (PDB ID : 4BAE) and MTB 3-deoxy-D-arabino-heptulosonat-7-phosphate synthase (PDB ID : 5E2L). The study showed that punicalin had the best interaction with MTB 3-deoxy-D-arabino-heptulosonat-7- phosphate synthase with binding energy -6,79 kcal/mole. Toxicity prediction showed that brevifolin was the safest substances among the other substances that contained in ethanol extract of pomegranate peel with binding energy -4,02 kcal/mol.
