The growth of microalgae and the amount of DHA obtained from it are affected by various factors; one of the factors is nitrogen source for the component of growth medium. The aim of this research is to carry out the best nitrogen source for microalgae growth medium that will produce the highest DHA level. This research uses three different nitrogen sources for microalgae growth medium such as bacteriological peptone, mycological peptone, and tryptone. The beginning of this research is the making of microalgae suspension and counted the microalgae cells by Total Plate Count (TPC). After that, the microalgae in the suspension are fermented for ten days using the liquid medium and the dried biomass is obtained from the fermentation process. Then the microalgae growth curve is made by plotting the dried biomass weight towards the fermentation duration. Furthermore, the DHA is extracted from the dried biomass that has the biggest weight through direct transesterification method. The DHA level is analyzed using gas chromatography with relative comparison towards the total amount of fatty acids in the extract. The results from this research are the best duration for fermentation of microalgae to produce the most biomass is seven days and the highest DHA level is obtained from the fermentation medium that used nitrogen source tryptone with the amount of 14.16 % relative to the total fatty acids in extract. text |
