Recently, one of the most widely studied new antimicrobial sources is the endophytic fungus. The aims of this research was to isolate endophytic fungi from marine-sponge obtained from Kepulauan Seribu and Pulau Samalona and investigate their antimicrobial activity against Escherichia coli ATCC8739, Staphylococcus aureus ATCC6538, and Candida albicans ATCC10231. Fungi isolation were conducted in YPD agar medium and each pure fungi was fermented for 21 days in YPD broth medium. The mycelium and broth culture were separated by using filtration method. Then, the mycelium was extracted by maceration and the supernatant was extracted by liquid extraction using ethyl acetate. Each extract was tested for their antimicrobial activity through determination of MIC and MBC/MFC. The results of this research were obtained 11 fungal isolates that were coded as SRMF1 until SRMF11 and all extracts showed activity against one or two bacterial test and just 6 extracts from 4 different fungal isolate that showed activity against C.albicans. The greatest antibacterial activity was showed by mycelium culture of SRMF1 extract (32µg/mL) against S.aureus, culture media of SRMF2 extract (16µg/mL) against E.coli, and mycelium culture of SRMF1 extract (32µg/mL) against C.albicans. The lowest MBC was 512µg/mL, showed by culture media of SRMF10 extract against S.aureus and mycelium culture of SRMF7 and SRMF9 extract against E.coli, while the lowest MFC was 64µg/mL, showed by mycelium culture of SRMF1 extract. Therefore, endophytic fungi that were Isolated from marinesponge have potential to be developed as an antimicrobial. text |
