limited for cigarettes production. Cigarette has negative effect on health. Empirically, tobacco is used
for many other purposes, one of them as an antiseptic. This study aims to prove and compare the
antimicrobial activity of unripe and ripe leaves of tobacco to confirm its use as antiseptic. Preparation
of unripe and ripe leaves extract was carried out by extraction of crude drug by reflux using ethanol
96%. The extract was then fractionated by liquid-liquid extraction method using n-hexane and ethyl
acetate. Antimicrobial activity of ethanol extracts, ethyl acetate fractions, and water fractions were
tested with paper disc diffusion method at concentrations 5, 10, and 20% against Escherichia coli,
Staphylococcus aureus, and Candida albicans. The result showed that the ethyl acetate fractions has
inhibitory effect on Escherichia coli and Staphylococcus aureus but not on Candida albicans. There was
no inhibitory effect observed in ethanol extracts. The water fraction of ripe leaves also have inhibitory
effect againts Staphylococcus aureus. This study showed that unripe and ripe tobacco leaves had
activity as an antimicrobial although not strong and there was no significant difference between
antimicrobial activity of unripe and ripe leaves. |
