Summary: | Pomegranate (Punica granatum L.) is one of the many plants that have various efficacy according
to several research recently. Some studies have proven that ethanolic extract of pomegranate
pericarp has activity as an antifungal but relatively still less developed. The aim of this research is
to observe antifungal activity on ethanol extract, n-hexane fraction, ethyl acetate fraction, and
water fraction of pomegranate pericarp to against Candida albicans ATCC 10231, Aspergillus niger
ATCC 16404 and Microsporum gypseum. The simplicia powder of pomegranate pericarp was
characterized and then extracted using reflux method with ethanol solvent then concentrated
into concentrated ethanol extract. The ethanol extract was fractionated by liquid-liquid extraction
using three solvents with increasing polarity, n-hexane, ethyl acetate, and water. The ethanol
extract, n-hexane fraction, ethyl acetate fraction, and water fraction carried out a preliminary test
to see the inhibitory diameter of the test fungi of each extract and fraction with disk diffusion
test. Furthermore, ethanol extract, ethyl acetate fraction, and water fraction were tested
qualitatively by microdilution method to determine the minimum inhibitory concentration (MIC)
value and determination the minimum fungicidal concentration (MFC) of each extract and
fraction of test fungi. In the Candida albicans fungus, pomegranate pericarp ethanol extract has
MIC and MFC value at ???ã?mL and 4096 ?ã???, a fraction of ethyl acetate at ????ã?mL and
4096 ?ã???, and water fraction at ???ã?mL and >4096 ?ã???whereas in Microsporum gypseum
fungus ethanol extract of pomegranate pericarp has MIC and value at ????ã?mL and 1024 ?ã???,
each ethyl acetate fraction at ????ã?mL, and water fraction at ????ã?mLand 1024 ?ã???.
Ethanol extract, ethyl acetate fraction, and water fraction of pomegranate pericarp contain
flavonoid, phenol, terpenoid, and quinone compounds. Ethanol extract, ethyl acetate fraction,
and water fraction has strong enough antifungal activity to the test fungi.
