Summary:In Indonesia the use of tobacco (Nicotiana tabacum L.) was already familiar as the main ingredient of cigarettes. In some studies, tobacco extract has been tested to have the benefit of being a natural insecticide. In addition, some studies has tested antibacterial activity in tobacco leaf on several leaf extracts of tobacco with different solvents. This research was aimed to isolate and identify the antibacterial compound from tobacco leaves against Staphylococcus aureus ATCC 6538 and Pseudomonas aeruginosa ATCC 9027. The tobacco leaves that have been drying dan grinding was extracted by reflux method using increasing polarity of solvent, n-hexane, ethyl acetate, and ethanol, then all of the extracts were monitored by thin layer chromatography (TLC) methode. Antibacterial activity test was performed by agar diffusion method, determination of minimum inhibitory concentration value (MIC) by microdilution, and also bioautography to determine target compound based on Rf that have inhibitory activity. The extract of ethyl acetate have highest activity based on the results of agar diffusion testing, microdilution, and bioautography was continued to the next stage. Results from bioautography showed extract component at Rf 0.66 in chloroform-ethyl acetate system (1: 1) gave antibacterial activity to both test bacteria. The ethyl acetate extract was fractioned using vacuum liquid chromatography method. Fractionation results were monitored using thin layer chromatography method and purified by preparative thin layer chromatography method. Compounds that have antibacterial activity were isolated and based on the characterization with specific visualization reagent, the isolate was a coumarin compound.
