Summary:Jackfruit is commonly consumed, however, the rag is still rarely used for its benefits. Empirically, jackfruit rag has been reported for its therapeutic usage such as reduction of cholesterol and blood sugar levels. A previous research reported that the rag contained alkaloids, flavonoids, saponins, tannins, phytosterols, glycosides, and phenols. The objective of this research was to isolate identity compound of jackfruit rag. The crude drug was extracted by reflux using ethanol. The extract was fractionated by liquid-liquid extraction and yielded ethanol, ethyl acetate, and n-hexane fractions. The chromatograms showed the presence of a dominant compound in the ethyl acetate fraction. The fraction was then subfractionated by radial chromatographic system, yielded 17 subfraction. One compound in subfraction 8 (subfraction A) appeared as a dominant spot under ultraviolet light of 254 nm and one compound in subfraction 11 (subfraction B) appeared as a blue fluorescent spot under ultraviolet light of 366 nm. Both compounds were isolated by preparative thin layer chromatography (TLC). The two isolates were confirmed as identity compounds. The Isolate A on GF254 TLC with developing system of ethyl acetate-n-hexane (1:1) having Rf 0.45, showed blue-green spots on anisaldehyde spray reagent, resulted a red-purple on Liebermann-Burchard reagent, showed maximum absorptions at 224 nm and 280 m, and having O-H, CH2, CH3, CH, and C = C aliphatic groups. The isolate B on GF254 TLC with developing system of toluene-acetone (1:2) having Rf 0.81, showed black spot with iron (III) chloride spray reagent and blue fluorescent under ultraviolet light of 366 nm with the citroborate spray reagent, showed a maximum absorption at 221 nm, and having O-H, CH2, CH3, CH, and C = C aliphatic.
