Summary:Spironolactone is a drug that commonly used to treat hypertension and edema conditions. Additionally, spironolactone also had an effect as antiandrogenic, potentially in the treatment of androgenic alopecia. Target og spironolactone for the treatment of androgenic alopecia is hair follicles. The application is used topically. It aims to reduce the dose of medication that can use systemic toxicity when administrated orally. One method used to maximize the delivery of spironolactone to the hair follicle is nano-sized lipid carrier, the NLC (Nanostructured Lipid Carrier). NLC is a typed of colloidal drug delivery systems, generated through the mixing of the solid lipid dan liquid lipid. This study aimed to develop formula of NLC spironolactone in order to fulfill the various parameters. NLC created by using high speed homogenization ultra turax and sonicated using a probe sonicator. The experiment began with the selection of a couple of solid lipid (glyceryl monostearate, cera alba, stearic acid, cetyl alcohol) and liquid lipid (oleic acid, virgin coconut oil and olive oil). The proceed with the selection of a surfactant (Tween 80, Plantacare, Tegocare). Spironolactone NLC evaluation include organoleptic, analysis of particle size, polydispersity index, the efficiency of entrapment and physical stability during storage. NLC Spironolactone was produced in 4,5% glyceryl monostearate, 1,5% oleic acid and 2% Plantacare with 1% active ingredient. Established composition of spironolactone loaded NLC was used for this study, resulting particle size of 137,87 ± 2,46 nm, polidispersity index of 0,29 ± 0,01, potential zeta of -2,13 mV and entrapment efficiency of 91,3%. NLC stability test result for several days showed increasing of particle size but still within the target range.
