Summary:Irbesartan is a drug belongs to Biopharmaceutical Classification System (BCS) class II with low solubility and high permeability. Irbesartan has two crystalline forms, form A and B. The purpose of this research was to investigate the influence of neat grinding and solvent-drop grinding with water and ethanol on polymorphic transformation of irbesartan. Grinding was carried out for 30, 60, and 120 minutes with and without solvent using mortar grinder. Samples were characterized by scanning electron microscope (SEM), powder x-ray diffractometry (PXRD), differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA). Raw material used was irbesartan form A. According to SEM, PXRD, DSC, and TGA analysis, neat grinding and solvent-drop grinding didn’t cause polymorphic transformation of irbesartan. But grinding caused reduction of crystallites sizes and crystal defect so that the crystallinity of irbesartan decreased while amorphous parts increased as grinding time increased. Solvent evaporation occured on samples of irbesartan grinded with solvent for 120 minutes on temperature around 70°C. Solvent-drop grinding caused the formation of aggregates, with larger sizes as the grinding time increased. Samples grinded with water formed bigger aggregates compared to the other grinded with ethanol.
