Cotrimoxazole is a drug consisting of sulfamethoxazole (SMZ) and trimetorpim (TMP). SMZ and TMP are practically insoluble in water and is a class II drug in biopharmaceutics classification system. Therefore, the bioavailability of cotrimoxazole is affected by the sollubility of cotrimoxazole itself. This study was aimed to improve the solubility of cotrimoxazole using solid dispersion. Preliminary test was performed by comparing the dissolution profile between two generic brands of cotrimokxazole with the dissolution profile of innovator product. The solid dispersion was prepared by dissolving technique using PVP K30. TMP and SMZ dissolved along with the PVP K30 in methanol until clear solution was obtained. Solid dispersion composed of cotrimoxazole and PVP in the ratio of 1:1/3; 1:1/2 and 1:1. Recrystallization process was allowed slowly at room temperature. Dissolution test for each composition and pure cotrimoxazole was performed in 3 different pH medium (pH1,2; pH 4,5; pH 6,8) using a dissolution apparatus type 1 (basket) at 100 rpm. Optimal amount of the carriers showed from a ratio of 1: 1/2. Results of dissolution test on three different medium that solubility of cotrimoxazole and solid dispersion cotrimoxazole was significantly different. text
