Summary:Treatment of disease with a combination of drugs is to enhance the effect or reduce adverse effect, one example of drug combination is losartan potassium and hydrochlorothiazide tablet. The combination of losartan potassium and hydrochlorothiazide is proven to have a significant increase in antihypertensive activity compared to its monotherapy. However, the components of drugs such as active pharmaceutical ingredients (API) and excipients can interact with each other. One of the interactions that can occur is physical interactions, which may affect the physicochemical properties of drugs. Physical interactions are not limited to occur between excipients, physical interactions can also occur between APIs. The physical interaction of losartan potassium and hydrochlorothiazide was studied further using crystallography-based methods such as hot stage microcopy, solvent-drop grinding, solvent evaporation method and powder X-ray diffraction (PXRD) analysis. Microscopic observation was carried using the polarized light microscope, equipped with hot stage and digital temperature control. Solvent-drop grinding methods were done to the mixture for 20, 40 and 60 minutes. Some of the solvent-grinded samples were taken for dissolution testing and PXRD analysis. Powder X-ray diffractogram showed no change of peaks of the grinded powder. The results showed that the physical interaction between losartan potassium and hydrochlorothiazide is a simple eutectic mixture.
