INSTITUT TEKNOLOGI BANDUNG https://digilib.itb.ac.id/gdl/view/44756 Fungi is heterotroph organism, unicellular or multicellular, and produces spores. Marine-derived fungi is fungi that grows and sporulates in marine environment. Marine-derived fungi has been developed as secondary metabolites producer that have antibacterial, antifungal, and antiviral activity. The aim of this research were to assay antibacterial activity of marine-derived fungi of culture media and mycelial extract of GK8 that produced from static fermentation, to get fractions of secondary metabolite, and to assay their antibacterial activity against Bacillus subtilis and Escherichia coli. Extraction was conducted by liquid-liquid extraction for culture media and maceration for mycelial using ethyl acetate. Fractionation was conducted by classic column liquid chromatography and TLC preparative. Antibacterial activity was tested by disk diffusion and bioautography. Extracts of culture media and mycelial of GK8 showed activity against Bacillus subtilis and Escherichia coli. The greatest activity was shown by culture media extract of GK8 against Bacillus subtilis (16.2 mm diameter inhibitory at level of 250 µg/disk). Majority fractions of culture media and mycelial of GK8 have good activity against Bacillus subtilis and low activity against Escherichia coli. The greatest activity was shown by fraction IV from culture media extract of GK8 against Bacillus subtilis (18.4 mm diameter inhibitory at level of 250 µg/disk). Bioautography assay showed that all of subfractions have antibacterial activity against Bacillus subtilis but did not show antibacterial activity against Escherichia coli. text
