In the case of fish, formaldehyde may be present from both intentional addition and natural formation due to enzymatic degradation of fish flesh. This study aims to obtain a method for determination of formaldehyde in fish flesh by means of spectrophotometry technique. The fish flesh samples were firstly homogenized and distillated under acidic condition. The distillate is then reacted with Nash reagent to form a yellow-colored solution spectrophotometrically measurable with maximum absorbance at 413 nm. Validation parameters such as linearity, limit of detection (LOD), limit of quantification (LOQ), accuracy and precision were determined. The method gave linear calibration curve in the concentration range of 2.5 to 6.0 ppm and having a regression equation of y = 0.1281x + 0.0046 with the correlation coefficient (r) of 0.999 and the relative procedural standard deviation (Vx0) of 0.88%. LOD and LOQ of the method were 0.14 and 0.38 ppm, respectively. In the precision test, the variance coefficient equals 3.97% for repeatability and 2.26% for intermediate precision. In the accuracy test, the recovery percentage of standard addition at the concentration levels of 0.5, 1.0 and 1.5 ppm were 97.81%, 98.05% and 99.16%. Meanwhile in the accuracy test for sample simulation results, the average recovery percentage was 50.29%, which should be taken into account for quantitative analysis. On testing 10 samples of fresh fish, none of the fish contains quantified formaldehyde. Based on overall results, it can be concluded that a method for determination of formaldehyde in fish flesh by means of spectrophotometry technique using Nash reagent was successfully obtained. text |
