mycobacterium. Determination of its potency in pharmaceutical formulation can be done with agar
diffusion bioassay. Agar diffusion has some design of assessment, including 5+1 with 5 dose of
standards and 1 dose of test solution, and 3+3 with 3 dose of standards and 3 dose of test solution.
The use of paper disc for agar diffusion bioassay has not been previously reported. Thus, this paper
reports validation of 5+1 and 3+3 agar diffusion bioassay using paper disc method to quantify
kanamycin sulfate in pharmaceutical samples. The strain Staphylococcus aureus ATCC 6538 was
used as the test microorganism, and the results of the proposed bioassay displayed high linearity,
precision, and accuracy. All the potency with 5+1 model result were found to be linear (r >0.99) in
the range of dose 6.4-15.6 µg, accurate (recovery 101.6%), and precise (coefficient of variance
inter-day 2.2% and intra-day 4.4%). All the potency with 3+3 model also found to be linear (r >0.99)
in the range of dose 3.2-12.8 µg, accurate (recovery 101.4%), and precise (coefficient of variance
inter-day 4.5% and intra-day 2.6%). The results validated the proposed microbiological assay, which
allows reliable quantitation of kanamycin sulfate in pharmaceutical samples.
