Summary:Diabetes mellitus is a metabolic disorder characterized by high levels of glucose in the blood. Madeira vine and cutleaf groundcherry had shown antidiabetic activity on the previous studies. The aim of this study is to evaluate antidiabetic activity of the combination of madeira vine and cutleaf groundcherry extracts compared to their single dose. Both crude drug were extracted by reflux method. The study involved glucose tolerance test using Wistar rat, antidiabetic test using alloxan-induced ddY mice, and histological test. For glucose tolerance test, rats were divided into six groups which are control group, 100 mg/kg bw madeira vine extract, 10 mg/kg bw cutleaf groundcherry extract, combination of madeira vine and cutleaf groundcherry extracts using 100 mg/kg bw and 10 mg/kg bw, and 50 mg/kg bw and 5 mg/kg bw, and glibenclamide. Measurement of glucose levels were conducted at baseline and 30, 60, 90, 120, and 150 minutes after administration of dextrose. For alloxan-induced antidiabetic test, mice were divided as same as glucose tolerance test with addition of a normal group. Mice in diabetic group were induced using alloxan monohydrate and given the test materials once daily for 21 days. On day 21, mice were sacrificed and the pancreas were isolated for histological tests. In glucose tolerance test, combination of madeira vine and cutleaf groundcherry extracts using half of their single dose gave the greatest percent reduction of glucose levels. In alloxan-induced antidiabetic test, the combination of madeira vine and cutleaf groundcherry using their single dose gave the greatest percent reduction of glucose levels. The result of histological test showed that the extracts can improve the regeneration of pancreatic beta cells. Using those methods, it can be concluded that the combination of madeira vine and cutleaf groundcherry extracts have better antidiabetic activity than their single dose.
