Papaya (Carica papaya L.) is one of plants that have antioxidant activity, in its leaves and ripe fruit. This study aims to determine antioxidant activity of papaya leaves extract through determination of IC50 DPPH (2,2-diphenyl-1-picrylhydrazyl), total phenolic, flavonoid, and carotenoid, and isolate an antioxidant compound from papaya leaves extract. Extraction was carried out by reflux using three different solvents with increasing polarity. Each extract was monitored by thin layer chromatography (TLC) Antioxidant activity, total phenolic, flavonoid and carotenoid content were done using UV-sinar tampak spectrophotometry. The highest antioxidant activity was shown by ethyl acetate extract (IC50 DPPH 0.619 ± 0.064 µg/mL). The highest total phenolic content was given by the ethanol extract (5.194 ± 0.003 g GAE/100 g), the highest total flavonoid by the ethyl acetate extract (24.478 ± 0.028 g QE/100 g), and the highest total carotenoid by n-hexane extract (26.881 ± 0.400 g BE/100 g). Fractionation of n-hexane extract by vacuum liquid chromatography (VLC) gave 20 fractions. Purification of fractions 5-7 were performed by preparative TLC. Purity test of band A by single development TLC using three different mobile phases and two dimensional TLC, indicated one spot. Isolate N positive with citroboric spray reagent, showed maximum wavelength at 274 nm and 362 nm, gave yellow color under UV ?366 nm. Antioxidant compound N was predicted to be a flavonol aglycone which has free OH at C-3 and contained many methoxyl groups. text
