The aim of this study was to determine which grade of black tea give the highest antibacterial activity against Streptococcus mutans. The 15 grades of black tea were extracted using maceration with methanol. All extracts were evaporated and tested for their antibacterial activity against Streptococcus mutans through minimum inhibitory concentration (MIC) using microdilution, minimum bactericidal concentration (MBC) by agar plate method, and bioautography methods. The methanol extract that gave strongest activity was fractionated by liquid-liquid extraction, with n-hexane, ethyl acetate, and water. All fractions were tested for antibacterial activity through MIC, MBC and bioautography method. Based on phytochemical screening, all 15 grades of black tea contained alkaloid, flavonoid, tannin, saponin and steroid/triterpenoid. PF II had the lowest MIC (625 µg/ml) compared to other grades and there was zone of inhibition zone at Rf 0.7 in bioautography result. Ethyl acetate fraction showed lowest MIC (1250 µg/ml) and was monitored with TLC which showed the presence flavonoid and catechin. The Rf value that showed zone of inhibition against Streptococcus mutans were at Rf 0.76 and Rf 0.3. From bioautography result, compound at Rf 0.76 was catechin. Among all grade of black tea, PF II had the strongest antibacterial properties against Streptococcus mutans. text |
