Summary:Gout is a metabolic disorder characterized by the increasing of uric acid concentration. The enzyme which plays an important role in the formation of uric acid is xanthine oxidase. The search for the treatment of gout as xanthine oxidase inhibitor can be obtained from plant sources. Akar wangi and buas-buas known to have strong antioxidant activity and traditionally used for anti-inflamation, rheumatism, and gout. The aims of this study were to determine the antioxidant activity and xanthine oxidase inhibitory from the root extract of Vetiveria zizanioides (L). Nash and the wood extract of Premna cordifolia L. that were macerated gradually with n-hexane, ethyl acetate, and methanol. The activity assays were performed using the DPPH radical and xanthine oxidase enzyme. The IC50 values of n-hexane, ethyl acetate and methanol extract of akar wangi were 988.16 ?g/mL, 391.43 ?g/mL, and 223.97 ?g/mL respectively. While the IC50 values of n-hexane, ethyl acetate and methanol extract of buas-buas were 862.80 ?g/mL, 456.14 ?g/mL, and 115.64 ?g/mL. The result showed that akar wangi and buas-buas methanol extract had the highest antioxidant activity. The inhibitory activity of xanthine oxidase that given by the akar wangi methanol extract had a weak characteristic with the IC50 value was 183.80 ?g/mL while buas-buas methanol extract had a strong characteristic with IC50 value was 93.22 ?g/mL.
