Summary: | Gout is a metabolic disorder that is characterized by attacks of acute arthritis that appears
repeatedly as a result of monosodium urate deposit in joint and cartilage. To resolve this disease,
there are some people who use the treatment options with traditional medicinal plants and some
others using a chemical drug, one of them is alopurinol. This research was done to proof and
verify the activity of xanthine oxidase inhibitor on the use of traditional medicinal plants that
thrive in Indonesia society. Plants were examined as many as 22 specieses. Activity of the samples
was done by comparing the absorbance of uric acid formed as a result of the reaction between
the substrate and enzyme xanthine oxidase with the extract and without it. If the absorbance of
uric acid formed in the test solution containing the extract is smaller than the test solution
without the extract, it is concluded that the extract has potential as inhibitor xanthine oxidase.
The results showed that the extract Muntingia calabura L., Litsea cubeba (Lour.) Pers. and
Plectranthus scutellarioides (L.) R.Br. has the highest activity, ie 67,87%, 67,79% and 66,68%, with
IC50 values respectively 17,42 µg/mL, 7,57 µg/mL and 10,68 µg/mL. Inhibitory xanthine oxidase
activity of alopurinol has percentage inhibiton 99,64% with IC50 value of 0,44 µg/mL. The highest
activity was shown by Muntingia calabura L. fruit extract.
