Summary:The uses of antibiotics combination are for a long time, included in the veterinary world. One antibiotic combination used is sulfadiazine and trimethoprim. Although the use of a combination of two antibiotics has been long established, not much researches to analyze these antibiotics combinations, including assay study of these antibiotics in blood and in other biological samples. Assay of both antibiotics used the method of high-performance liquid chromatography (HPLC). Optimization method is done by replacing the mobile phase composition and wavelength of the detector UV-Vis spectrophotometer. This HPLC method system used a C18 column (150 mm x 4.6 mm). Then the detail: flow rate 1 ???ð???????± ?» ð?ü±®?ð????????­ð?± ?í ?± ®?????ð?ð???» acetonitrile: 1.27% glacial acetic acid: triethylamine (14 : 85.85 : 0.015), and the detector UV-Vis spectrophotometer at a wavelength of 270 nm. Sulfadiazine and trimethoprim had respective retention time of 3.4 and 4.6 minutes. Sulfadiazine had a linearity value of 0.9998 with a detection ?ð?ð??» ?????ã ?ã??  ?¸   ?? ??ð»ð® ?ð???ð?ð??» ?????ã????ð?±?í???ð?í ¸   ?ð?± ?ð??? ??± ?» ???????ð?í   ¸±?±®?ð???ð?ð??» ?????ã??  ?¸   ?? ??ð»ð® ?ð???ð?ð??» ?????ã???&±?í?¸ validation has met the requirements of precision and accuracy, giving the value of the coefficient of variance and the % error does not exceed 15%. The sample preparation technique used protein precipitation with acetonitrile solvent and produced good recoveries and precise. Samples in the plasma could last up to 34 days, shown by the stability test using statistic t-student test. Proven methods could be used to determine the levels of sulfadiazine and trimethoprim in the blood to determine pharmacokinetics profile of chicken by the drinking water route.
