To prevent the liver injury, hepatoprotective is required. Ethanolic extract of Mulberry leaves and Binahong leaves on the previous studies had shown hepatoprotective activity, but there has been no research that observes the effect of combination yet. Therefore, this study aims to evaluate hepatoprotective activity of combined ethanolic extracts of Mulberry leaves (DM) and Binahong leaves (DB) that compared with their respective single dose against paracetamol-induced hepatotoxicity in female Wistar rats. Both crude drugs of DM and DB extracted by reflux method using ethanol as solvent. Rats were divided into 7 groups which are normal group, positive control, Silymarin 100 mg/kg bw, DM extract 200 mg/kg bw, DB extract 100 mg/kg bw, Combination A (DM 200 mg/kg bw ?DB 100 mg/kg bw), and Combination B (DM 100 mg/kg bw ?DB 50 mg/kg bw). Rat were given the test material and induction using paracetamol 900 mg/kg bw for 14 days. Determination parameters of the ALT and AST test done on day-0, 7, and 14. On day 14, the rat was sacrificed and liver organ was isolated to determine organ index and histology. The results on the 7th day showed that the whole group test has values of ALT which significantly lower than positive control, whereas the activity of AST indicate only Silymarin group and Combination B which had values of AST significantly lower than positive control. On day 14, , the whole group test has values of ALT and AST which significantly lower than positive control. The result of organ index showed that all groups had not significantly different. Among the test groups, liver histology test which had the minimal damage showed on Combination B group. Using those methods, it can be concluded that Combination B (DM 100 mg/ kg bw ?DB 50 mg/kg bw) showed hepatoprotective activity better than their respective single dose and comparable to Silymarin 100 mg/kg bw. text
