HBx!mutant!in!patients!with!chronic!hepatitis!B,!cirrhosis!and!HCC.!HBx!K130M/V131I!has!been! studied!extensively!for!its!association!with!HCC.!However,!the!effect!of!HBx!T118N!and!K130M!in! liver!carcinoma!has!never!been!studied!before.!The!aim!of!this!study!was!to!determine!the!effect! of!HBx!T118N!and!K130M!expression!in!HepG2!cells!using!colony!formation!assay!and!the!results! were! used! as! suggestion! for! counseling.! Plasmids! pD609_HBx_wt,! pD609_HBx_T118N! and! pD609_HBx_K130M! were! first! isolated,! confirmed! and! then! transfected! into! HepG2! cells.! Succesful! transfection! was! detected! by! measuring! HBx! mRNA! expression! using! Reverse, Transcriptase0Polymerase, Chain, Reaction! (RTZPCR).! Selection! of! HepG2! cells! using! various! puromycin!showed!optimum!concentration!at!0,2!?g/ml.!Colony!formation!assay!was!performed! by!growing!transfected!cells!in!media!with!0,2!?g/ml!puromycin!for!14!days.!The!result!showed! that!number!of!colonies!formed!upon!HBx!T118N!and!K130M!expression!were!less!than!wildZtype!
