Summary: | Cardiovascular disease is one of the major causes of death in the world. Hypertension is one of
the most significant risk factors of cardiovascular disease. Mulberry leaves and celery leaves have
been used traditionally for hypertension. This study aims to determine the blood pressure
lowering effect of the combination of the mulberry (Morus nigra L.) leaves ethanol extract and
celery (Apium graveolens L.) leaves aquoeus extract in dexamethasone-induced hypertension in
male Wistar rats. The activity of blood pressure lowering effect was determined by testing the
combination of the extract which was administered orally on day 8 to day 14 in dexamethasoneinduced hypertension rats. Blood pressure were measured using a tail cuff (CODA®) on day 0,
after induction at day 7, and one hour after treatment on day 8, day 10, and day 14. Sistolic blood
pressure (SBP) increased significantly (p<0,05) in all groups at day 7. The mean increase of SBP
were 46.68 mmHg. This suggested that the animal models of hypertension had been formed.
Upon the treatment with amlodipine at a dose of 0.45 mg/kg bw rat, celery at a dose of 54 mg/kg
bw rat, and the combination of ½:½ of mulberry and celery dosage, SBP could be decreased
significantly compared to positive group after 3 day of treatment with reduction to 26.70 mmHg,
36.15 mmHg and 20.45 mmHg respectively. The mulberry at a dose of 400 mg/kg bw rat and
combination of 1:1 of mulberry and celery dosage showed a significant decrease of SBP after 7
day of treatment with the reduction to 41.80 mmHg dan 53.26 mmHg respectively. Diastolic
blood pressure (DBP) could be decreased significantly compared to positive group after 3 day of
treatment at the comparison group, celery, combination 1:1, and the combination ½:½ grup with
reduction to 20.80 mmHg, 27.75 mmHg, 20.1 mmHg, and 20.3 mmHg respectively. The mulberry
group showed a significant decrease of DBP after 7 day of treatment with the reduction to 35.75
mmHg. From this study it can be concluded that the combination of ethanol extract of mulberry
leaves and water extract of celery leaves with a ratio of a dose of 1:1 (mulberry extract at a dose
of 400 mg/kg bw rat and celery extract at a dose of 54 mg/kg bw rat) and a dose of ½: ½ (mulberry
extract at a dose of 200 mg/kg bw rat and celery extract at a dose of 27 mg/kg bw rat) showed the
blood pressure lowering effect. The combination group at a dose ½:½ showed the optimum blood
pressure lowering effect.
