Summary:Indonesia is an archipelago (most of its territory is ocean) with a tropical climate so that the diversity of Indonesian marine microalgae is very high. The diversity of the Indonesian marine microalgae needs to be protected. One of the protection steps is spesies identification. This study aimed to identifY a microalgae specimen isolated from the Indonesian Thousand Islands through genetic approaches. The microalgae sample was analyzed under a light microscope to investigate the cell shape and color. Genetical analysis was performed on ISS rONA-V 4 and LSU 02/03 markers. The DNA markers were amplified by PCR tcchniyue, and nucleotide scyucncc of the amplicon were determined by sequencing technique. The nucleotide sequences were then analyzed using several software, such as: seqman, BioEdit, BLAST (NCBI). and MEGA7. Results showed that the microalgae KSI was round in shape and brown in color. The genetic analysis classified this microalgae in to genus Cyclotella, with the highest identity (i.e. I 00% based on ISS rONA - V 4; 99,52% based on LSU 02/03 ) to the species Cyc/otella striata.
