INSTITUT TEKNOLOGI BANDUNG https://digilib.itb.ac.id/gdl/view/45370 Indonesia as a part of Southeast Asia region has the highest incidence of mortality caused by typhoid fever. Based on previous ethnopharmaca surveys, candlenut stem bark [Aleurites moluccana (L.) Willd] used by East Borneo peoples for typhoid treatment. Screening antibacterial of methanol extracts from stem bark of A. moluccana using diffusion method showed good inhibition against Salmonella typhi. Because of that, it would be interesting for isolating active compounds from A. moluccana stem bark that have antibacterial activity against Salmonella typhi. Methods: Extraction was done by maceration using methanol. Fractionation was performed with liquid-liquid extraction method using n-hexane and ethyl acetate, then it was continued using vacuum liquid chromatography (VLC) and chromatotron. The separation process was monitored and selected based on the antibacterial activity againts Salmonella typhi with bioautography TLC method. Purification was done using recrystallization, the result was monitored using 2- dimensional TLC. Microdilution test was used to obtain the MIC values of isolates. Characterization of isolates were done using UV-Vis spectrophotometer, 1D-NMR ( 1 H-NMR, NOE, 13 C-NMR, and DEPT) and 2D-NMR (HSQC and HMBC) spectroscopy. Results: Isolate (1) is white crystal that was obtained from fraction A of KCV results of the ethyl acetate extract. Isolate (2) is yellowish crystal obtained from fraction H2 of KCV and chromatotron results, this fraction had antibacterial activity respectively against Salmonella typhi. MIC values of isolates 1 and 2 were 500 ppm and 250 ppm. Based on spectroscopic data and previous studies, the isolated compounds were 3-acetylaleuritolic acid (1) and scopoletin (2). text
