Summary:Nanocarrier with PLGA polymer base often used as an effective carrier for drug delivery to cell. PLGA nanoparticle is easily recognized by the immune system and eliminated from circulation system because of its hydrophobic characteristic. It allows opsonisation and elimination from reticuloendothelial system (RES). Modification in the surface of PLGA nanoparticle to prevent opsonisation resulting in long-circulating nanoparticle can be done with hydophylic polymer, for example poloxamer. Nanocarrier also have to be safe and have selective toxicity. In vitro cytotoxicity test with cell culture is used for cytotoxicity screening for drug compound. This study aims to determine the cytotoxicity profile from nanocarrier containing artemisinin with PLGA-poloxamer polymer combination. Nanocarrier containing artemisinin was made with nanoprecipitation method. Cytotoxicity test was done with MTT method on Vero cell. Its absorbation was measure with microplate reader, then compared to cell control absorbation, and used to measure the percentage of living cells. The result showed that nanocarrier containing artemisinin with poloxamer 10%, PLGA 1%, and ART 30 ppm formula have the highest cytotoxicity profile (7,545% living cells). The one with poloxamer 3%, PLGA 0,2%, and ART 10 ppm formula showed lowest cytotoxicity profile (39,057% living cells). And the one with 5%, PLGA 0,2%, and ART 10 formula showed low cytotoxicity and the longest-circulating profile (30,66% living cells). The conclusion of this study is that formula with poloxamer 5%, PLGA 0,2%, and ART 10 ppm give a low and longest-circulating cytotoxicity profile, the formula poloxamer 3%, PLGA 0,2%, and ART 10 ppm showed lowest cytotoxicity profile, and formula poloxamer 10%, PLGA 1%, and ART 30 ppm showed highest cytotoxicity profile.
